Biological Background: AKT1 Function and Localization

  • AKT1, also known as RAC-alpha serine/threonine-protein kinase, Protein kinase B (PKB), or Proto-oncogene c-Akt, is a member of the AGC kinase family.
  • AKT1 is one of three closely related AKT kinases (AKT1, AKT2, AKT3) that regulate key cellular processes including metabolism, proliferation, cell survival, growth, and angiogenesis.
    Related references: PMID:11882383 PMID:15526160 PMID:15861136
  • It serves as a central downstream effector of phosphatidylinositol 3-kinase (PI3K) signaling, mediating the effects of growth factors such as insulin, IGF1, EGF, and PDGF.
    Related references: PMID:12176338 PMID:12964941
  • AKT1 phosphorylates a broad spectrum of substrates (over 100 reported) to influence diverse pathways: for example, it inactivates GSK3 to promote glycogen synthesis, phosphorylates FOXO transcription factors leading to their cytoplasmic retention and inhibition of pro-apoptotic gene expression, activates mTORC1 via TSC2 phosphorylation, and phosphorylates BAD to suppress apoptosis.
    Related references: PMID:11882383 PMID:15526160 PMID:21432781
  • Subcellular localization: AKT1 is found in the cytoplasm, nucleus, cell membrane, and mitochondrial intermembrane space, reflecting its involvement in multiple signaling compartments.
  • Tissue expression: ubiquitously expressed in all human cell types analyzed; increased protein levels observed in prostate and breast cancers during progression.
  • Post-translational modifications: AKT1 undergoes phosphorylation (activation), acetylation, ubiquitination, and disulfide bond formation, which regulate its activity and stability.

Experimental Guidance and Technical Tips

  • The immunogen used is a synthetic peptide spanning residues 381–480 of human AKT1 (C-terminal region). For Western blotting, expect a specific band around 56 kDa; however, note that AKT1 has multiple phosphorylation and other modifications that may alter its electrophoretic mobility.
  • For immunohistochemistry (IHC-P), antigen retrieval optimization is recommended; test on known positive tissues (e.g., human prostate or breast cancer sections) and include appropriate controls.
  • In immunoprecipitation (IP) and ELISA, the rabbit monoclonal format may provide high specificity and batch-to-batch consistency; validate the antibody in the relevant sample matrix.
  • Cross-reactivity with mouse and rat AKT1 is indicated, but endogenous expression levels vary by tissue; consider verifying reactivity in your specific experimental system.
