- AK5 (adenylate kinase isoenzyme 5, also known as ATP-AMP transphosphorylase 5) is a nucleoside monophosphate kinase that catalyzes the reversible transfer of the terminal phosphate group between nucleoside triphosphates and monophosphates.
- The enzyme is active on AMP and dAMP with ATP as the phosphate donor; when GTP is used as donor, it phosphorylates AMP, CMP, and to a small extent dCMP. It also exhibits broad nucleoside diphosphate kinase activity.
- Encoded by the AK5 gene, alternative splicing produces multiple isoforms of this kinase.
- AK5 is localized predominantly in the cytoplasm.
- Expression of AK5 is brain-specific, making it a candidate marker for studying neuronal function and neurological disorders.
- The protein features ATP-binding and nucleotide-binding domains, typical of the adenylate kinase family, and is classified as a transferase.

- The immunogen corresponds to residues 150–370 of human AK5, which is present in all known isoforms; consider verifying isoform coverage if studying specific splice variants.
- For Western blot, use brain-derived lysates (e.g., whole brain, cerebellum) as positive controls; a band around 63 kDa is expected, but validate in your sample system.
- IHC-P on brain tissue sections may require optimization of antigen retrieval; testing on both human and rodent tissues is advisable given the cross-reactivity profile.
- When performing ELISA, use appropriate coating conditions and confirm specificity with recombinant AK5 or peptide competition where possible.
