Biological Background: AGR2 Function and Localization
- AGR2 (anterior gradient protein 2 homolog), also designated HPC8 or XAG-2 homolog, is a secreted and endoplasmic reticulum-resident protein involved in tissue homeostasis and cancer.
- It is essential for post-transcriptional synthesis and secretion of MUC2, contributing to mucosal barrier integrity in the intestine (By similarity).
- As a proto-oncogene, AGR2 promotes cell migration, differentiation, growth, and enhances cell adhesion (PubMed:23274113). Related references: PMID:23274113
- AGR2 localizes predominantly to the endoplasmic reticulum but is also secreted into the extracellular space.
- Exhibits strong expression in trachea, lung, stomach, colon, prostate, and small intestine; weaker expression in pituitary, salivary gland, mammary gland, bladder, appendix, ovary, fetal lung, uterus, pancreas, kidney, fetal kidney, testis, placenta, thyroid, and ER-positive breast cancer cell lines.
- Contains an N-terminal signal peptide and forms a disulfide bond, consistent with its processing through the secretory pathway.
Experimental Guidance and Technical Tips
- The polyclonal antibody was raised against amino acids 21–175 of mature human AGR2; for immunohistochemistry on formalin-fixed paraffin-embedded tissue, heat-induced antigen retrieval in citrate or EDTA buffer may improve staining.
- In Western blot, expect a band around 20 kDa; however, potential dimerization or glycosylation could produce additional bands—verify with appropriate controls in your sample system.
- For immunofluorescence/ICC, permeabilize cells with 0.1–0.5% Triton X-100 to detect ER-localized AGR2; paraformaldehyde fixation is recommended.
- The antibody shows cross-reactivity with mouse and rat AGR2; confirm sequence identity if precise quantification across species is needed.
- For ELISA, consider testing a range of coating antigen concentrations to optimize signal.
