Biological Background: ADPRM Function and Localization
- ADPRM (also known as MDS006, C17orf48, NBLA03831) is a manganese-dependent ADP-ribose/CDP-alcohol diphosphatase (ADPRibase-Mn, CDP-choline phosphohydrolase).
- It hydrolyzes ADP-ribose, ADP, CDP-glycerol, CDP-choline, CDP-ethanolamine, and cyclic ADP-ribose (cADPR), but not other non-reducing ADP-sugars or CDP-glucose. Related references: PMID:25692488
- By similarity, ADPRM may participate in immune cell signaling, as its substrate ADP-ribose acts as a second messenger activating TRPM2 channels in response to oxidative/nitrosative stress.
- The protein undergoes acetylation and alternative splicing, and contains metal-binding sites for manganese and zinc (UniProt Keywords: Acetylation, Alternative splicing, Metal-binding, Zinc).
- Predicted molecular weight is approximately 39.5 kDa (UniProt), with an observed band at 40 kDa under reducing conditions (product specification).
Experimental Guidance and Technical Tips
- The immunogen (amino acids 203-342 of human ADPRM) targets the C-terminal region; consider validating specificity across known isoforms and using positive controls.
- For Western blotting, a band near 40 kDa is expected; optimize detection in human, mouse, and rat lysates and titrate the antibody for optimal signal-to-noise ratio.
- For ELISA applications, consider testing both direct and indirect formats and include cross-reactivity controls for related ADP-ribose metabolizing enzymes.
- Because ADPRM is a hydrolase, functional complementation with activity assays may help correlate protein levels with enzymatic function.
