Biological Background: ADAM9 Function and Localization
- ADAM9 is a disintegrin and metalloproteinase domain-containing protein 9, also known as meltrin-gamma, MCMP, MDC9, or CORD9. It is a member of the ADAM family and plays key roles in cell adhesion and proteolysis.
- Functions as a metalloprotease that cleaves and releases molecules important for tumorigenesis and angiogenesis, including TEK, KDR, EPHB4, CD40, VCAM1, and CDH5.
- May act as an alpha-secretase for amyloid precursor protein (APP), contributing to non-amyloidogenic processing.
- Mediates cell-cell and cell-matrix interactions and may regulate cell motility through interactions with integrins.
- Subcellular localization includes the cell membrane and also secreted forms.
- Widely expressed in various tissues, with isoform 2 highly expressed in liver and heart. Also expressed in chondrocytes.
- Post-translational modifications include glycosylation, disulfide bond formation, and phosphorylation; contains a zinc-binding motif for its protease activity.
- Associated with cone-rod dystrophy through alternative splicing and mutations.
Experimental Guidance and Technical Tips
- The immunogen spans the N-terminal region (aa 29-210) of human ADAM9, which includes the prodomain and part of the metalloprotease domain. For Western blotting, consider using reducing conditions and validate the full-length band at ~91 kDa as well as possible processed forms.
- For immunofluorescence (IF/ICC), expect membrane localization; appropriate fixation and permeabilization may be needed for optimal detection.
- ELISA applications can be optimized by coating with the recombinant immunogen or native samples. Ensure to validate the antibody in your specific experimental system, as performance may vary.
