Biological Background: ACSL5 Function and Localization
- ACSL5, also known as ACS5 or FACL5, encodes long-chain-fatty-acid--CoA ligase 5, which catalyzes the ATP-dependent activation of long-chain fatty acids into acyl-CoA thioesters for both cellular lipid synthesis and mitochondrial β-oxidation. Related references: PMID:17681178, PMID:22633490, PMID:24269233
- It exhibits a broad substrate range with a preference for C16-C18 unsaturated fatty acids (By similarity).
- ACSL5 may direct exogenous fatty acids into triacylglycerol storage and may also stimulate fatty acid oxidation (By similarity).
- At the villus tip of the small intestine, ACSL5 sensitizes epithelial cells to TRAIL-mediated apoptosis, suggesting a role in epithelial turnover.
- It has been implicated in the survival of glioma cells, indicating a potential function in cancer biology.
- The protein localizes to the mitochondrial outer membrane, endoplasmic reticulum membrane, and cell membrane, consistent with its roles in lipid metabolism and signaling.
- ACSL5 undergoes alternative splicing, acetylation, and phosphorylation, which may regulate its activity and subcellular distribution.
Experimental Guidance and Technical Tips
- The immunogen spans the C-terminal region (aa 500–683), which may allow detection of multiple ACSL5 splice variants if the epitope is conserved. Verify in relevant samples.
- For Western blotting, expect a band near 76 kDa. Include appropriate positive and negative controls to confirm specificity.
- In immunofluorescence experiments, optimize permeabilization conditions to account for the protein's membrane‐associated localization.
- For IHC-P, antigen retrieval and detection protocols should be individually optimized. Validate staining patterns in tissue sections with known ACSL5 expression.
- This monoclonal antibody can be used in ELISA; consider pairing with a suitable detection antibody for quantitative assays.
