Biological Background: ACSL4 Function and Localization
- ACSL4 encodes long-chain-fatty-acid--CoA ligase 4, also known as arachidonate--CoA ligase or LACS4, a key enzyme in lipid metabolism.
- It catalyzes the ATP-dependent formation of long-chain acyl-CoA from long-chain fatty acids, essential for both lipid synthesis and mitochondrial β-oxidation. Related references: PMID:21242590, PMID:22633490, PMID:24269233
- ACSL4 preferentially activates arachidonate and eicosapentaenoate, and also converts epoxyeicosatrienoic acids (EETs) with a specificity order of 8,9-EET > 14,15-EET > 5,6-EET > 11,12-EET. Related references: PMID:21242590
- It modulates glucose-stimulated insulin secretion by regulating levels of unesterified EETs (By similarity).
- ACSL4 regulates prostaglandin E2 secretion, linking it to inflammatory pathways. Related references: PMID:21242590
- Acting as a critical activator of ferroptosis, ACSL4 converts polyunsaturated fatty acids (e.g., arachidonate and adrenate) into acyl-CoA derivatives, providing primary substrates for lipid peroxidation that trigger ferroptotic cell death. Related references: PMID:27842070, PMID:35027735, PMID:38720107
- The protein localizes to the mitochondrial outer membrane, endoplasmic reticulum membrane, and cell membrane.
- Mutations in ACSL4 are linked to X-linked intellectual disability (XLID) and other disorders, consistent with its aliases MRX63/MRX68/XLID63.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant fusion protein spanning amino acids 1-280 of human ACSL4 (NP_004449.1). This N-terminal region is expected to elicit antibodies that recognize the full-length protein and possibly some splice variants. Consider verifying reactivity against your target isoforms.
- This antibody is suitable for Western blot (WB), where the predicted molecular weight is 79 kDa. Run appropriate positive controls (e.g., tissues or cell lines with known ACSL4 expression) and confirm band size.
- For immunohistochemistry (paraffin sections, IHC-P), antigen retrieval conditions may require optimization. Use known positive tissue sections as controls.
- In immunofluorescence/immunocytochemistry (IF/ICC), validate antibody specificity with appropriate controls (e.g., siRNA knockdown, blocking peptide, or knockout cell lines).
- The antibody has demonstrated cross-reactivity with mouse and rat samples, but we recommend validating reactivity and optimal dilutions in your own experimental system.
