Biological Background: Histone H4 Function and Localization
Histone H4 is a core component of the nucleosome, which packages DNA into chromatin and regulates access to genetic information. It is essential for transcription, DNA repair, replication, and chromosomal stability. The dynamic post-translational modifications (PTMs) of histones, collectively called the "histone code," play a key role in epigenetic regulation.
Acetylation of histone H4, particularly at lysine residues such as K8, is a well-established mark associated with open chromatin and active gene transcription. The acetylation status is dynamically regulated by histone acetyltransferases (HATs) and deacetylases (HDACs).
Subcellularly, histone H4 is primarily localized in the nucleus and on chromosomes. It is subject to numerous types of PTMs including acetylation, methylation, phosphorylation, and ubiquitination, which modulate its function.

Experimental Guidance and Technical Tips
- For Western blot (WB), detect endogenous acetyl-histone H4 at approximately 11 kDa. Use acid-extracted histones or sonicated nuclear lysates. Optimize antibody dilution (suggest starting at 0.5-2 µg/mL) and use appropriate loading controls (e.g., total histone H4 or H3).
- For Immunoprecipitation (IP), cross-linked chromatin preparation methods are recommended to preserve acetylation marks. The antibody may be suitable for native ChIP; consider using quantitative PCR or sequencing for analysis.
- For IHC-P, antigen retrieval with citrate buffer (pH 6.0) may improve staining. Include positive and negative control tissues with known acetylation levels.
- For ELISA, this antibody may be used for peptide- or histone-based detection assays. Validate specificity using non-acetylated peptide.
