Biological Background: Histone H4 Function and Localization
Histone H4 is one of the four core histone proteins that form the nucleosome, the fundamental repeating unit of chromatin. Along with histones H2A, H2B, and H3, it wraps and compacts DNA, thereby regulating access to the genetic material. This compaction is essential for proper chromosome segregation during cell division and for the spatial organization of the genome within the nucleus. Histone H4 plays a central role in DNA-templated processes such as transcription, replication, and repair by controlling DNA accessibility through a complex array of post-translational modifications, collectively known as the histone code.

The N-terminal tail of histone H4 is subject to extensive post-translational modifications, including acetylation, methylation, phosphorylation, and others. Acetylation at lysine 12 (H4K12ac; corresponding to residue 13 of the mature protein) is a well-characterized histone mark associated with transcriptionally active chromatin and is recognized by various bromodomain-containing proteins. This modification is dynamically regulated by histone acetyltransferases (HATs) and deacetylases (HDACs), and its presence at specific genomic loci correlates with gene activation and open chromatin states.

Histone H4 is predominantly localized in the nucleus and is tightly associated with chromosomal DNA. In certain contexts, such as during spermatogenesis, acetylated histone H4 becomes enriched in the nucleus of elongating spermatids, suggesting a role in chromatin remodeling during germ cell development. The protein is ubiquitously expressed across tissues, reflecting its fundamental role in chromatin structure.

Experimental Guidance and Technical Tips
- For chromatin immunoprecipitation (ChIP) experiments, consider optimizing crosslinking time and sonication conditions to shear chromatin to a size range of 200–500 bp, as histone–DNA interactions are robust.
- When performing western blotting, enrichment of histones by acid extraction or histone purification kits can improve signal. Keep samples on ice and add appropriate histone deacetylase inhibitors if acetylation levels need to be preserved.
- This antibody is predicted to have wide cross-reactivity across species due to the highly conserved sequence of histone H4. Validate the antibody in the species and tissue system of interest, as acetylation patterns may vary.
- For immunohistochemistry (IHC-P), antigen retrieval with citrate buffer and careful optimization of antibody concentration are recommended to achieve specific nuclear staining.
