Experimental Guidance and Technical Tips
- The immunogen is a synthetic acetylated peptide around K18 of human Histone H3. The resulting rabbit monoclonal antibody is expected to specifically recognize acetylated H3K18, with no cross-reaction to unmodified K18 or other acetylation sites. Validate specificity using both acetylated and non-acetylated peptide competition assays or by treating cells with HDAC inhibitors.
- For Western blotting (WB), a single band at ~16 kDa is anticipated. Use appropriate loading controls (e.g., total Histone H3 or H3 C-terminal antibodies) to normalize acetylation levels. Acetylation signals may be enhanced by treating cells with deacetylase inhibitors such as trichostatin A (TSA) or sodium butyrate.
- For immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), optimize antigen retrieval and permeabilization conditions. Endogenous peroxidase quenching is recommended for IHC-P. Validate staining by comparing with isotype controls.
- For chromatin immunoprecipitation (ChIP), ensure cross-linking conditions are optimized for histone modifications. Use appropriate positive and negative control loci (e.g., actively transcribed genes vs. silenced loci). Enrichment efficiency can be verified by qPCR.
- Due to wide predicted cross-reactivity (human, mouse, rat, and others), confirm reactivity in the species of interest before proceeding with large-scale experiments. Sequence alignment shows high conservation of the region surrounding K18 across vertebrates.
- For ELISA and dot blot (DB) applications, the antibody can be used to detect acetylated H3K18 peptides or recombinant histones. Consider using a biotinylated secondary antibody and streptavidin-HRP for signal amplification.
