Biological Background: Histone H2B Function and Localization

Histone H2B is a core component of the nucleosome, the fundamental unit of chromatin. Together with histones H2A, H3, and H4, it wraps and compacts DNA, thereby regulating DNA accessibility. Post-translational modifications (PTMs) of histones, including acetylation, methylation, phosphorylation, and ubiquitination, constitute the "histone code" that dictates chromatin dynamics and gene expression. Experimental Guidance and Technical Tips

  • This antibody was raised against a synthetic acetylated peptide corresponding to K5 of human histone H2B. It is expected to specifically recognize the acetylated form; validate in your sample system using appropriate positive and negative controls (e.g., samples treated with HDAC inhibitors vs. untreated).
  • For Western blot (WB), consider using a nuclear-enriched lysate and standard SDS-PAGE conditions. A band around 14 kDa (the approximate molecular weight of histone H2B) should be detected.
  • For IHC-P, IF/ICC, and ELISA, optimize antibody dilution and include antigen retrieval steps as needed. Since the immunogen is a short peptide, antigen retrieval may be critical for IHC.
  • The antibody is predicted to cross-react widely across human, mouse, rat, and other species due to the highly conserved sequence around K5. However, cross-reactivity should be confirmed experimentally in each species of interest.
