Biological Background: Histone H2B Function and Localization
Histone H2B is a core component of the nucleosome, the fundamental repeating unit of chromatin. Together with histones H2A, H3, and H4, it wraps and compacts DNA, thereby regulating DNA accessibility to cellular machineries. Histone H2B plays a central role in transcription regulation, DNA repair, DNA replication, and chromosomal stability. DNA accessibility is regulated through a complex set of post-translational modifications (PTMs), collectively known as the histone code, which includes acetylation, methylation, phosphorylation, and others. The two major human H2B variants (SWISS P62807 and Q16778) share nearly identical sequences and are encoded by different gene clusters. Related references: UniProtKB P62807, UniProtKB Q16778.
In addition to its nuclear roles, Histone H2B exhibits broad antibacterial activity and may contribute to the antimicrobial barrier of the colonic epithelium and the bactericidal activity of amniotic fluid.
Histone H2B localizes predominantly to the nucleus and is tightly associated with chromosomes. It is widely expressed across tissues, reflecting its fundamental role in genome organization. Related references: UniProtKB P62807, UniProtKB Q16778.
Acetylation at lysine 20 (K20) is a specific PTM that can influence chromatin structure and gene expression. Mass spectrometry studies have identified numerous acetylation sites on H2B, including K20 (reported as K21 in some alignments). This antibody specifically recognizes the acetylated form of K20, enabling precise monitoring of this modification in chromatin biology and epigenetic research. Experimental Guidance and Technical Tips
- This polyclonal antibody was raised against a synthetic acetylated peptide corresponding to lysine 20 of human Histone H2B. It is expected to specifically recognize acetylated H2B‑K20 in relevant assays.
- For Western blotting (WB), consider using nuclear extracts or whole cell lysates from human, mouse, or rat samples. The predicted molecular weight is approximately 14 kDa. A recommended starting dilution should be empirically determined and validated in the relevant sample system.
- In ELISA, the antibody can be used as a capture or detection reagent. Pair with an appropriate total H2B antibody for normalization. Validate specificity using acetylated and non‑acetylated peptide competitors.
- For chromatin immunoprecipitation (ChIP), optimize cross‑linking and chromatin shearing conditions for your cell type. Include an anti‑Histone H3 or total H2B antibody as a positive control, and an IgG isotype control to assess non‑specific binding.
- Additional applications may be possible; always use appropriate controls and validate performance in your experimental context.
