Biological Background: ABHD12 Function and Localization
- ABHD12 is a lysophosphatidylserine (LPS) lipase that mediates hydrolysis of lysophosphatidylserine, a signaling lipid regulating immunological and neurological processes. It functions as the major LPS lipase in the brain, playing a critical role in the central nervous system. Related references: PMID:25290914, PMID:30237167, PMID:30420694
- Also known as 2-arachidonoylglycerol hydrolase ABHD12, monoacylglycerol lipase ABHD12, and oxidized phosphatidylserine lipase ABHD12, reflecting its multi-substrate specificity. It belongs to the abhydrolase domain-containing protein family (hABHD12).
- ABHD12 exhibits monoacylglycerol (MAG) lipase activity, hydrolyzing 2-arachidonoylglycerol (2-AG) and thereby modulating endocannabinoid signaling pathways. Related references: PMID:22969151, PMID:24027063
- The enzyme prefers lysophosphatidylserine substrates with long and very long chain acyl chains, showing strong preference for very-long-chain lipids due to improved catalysis rather than substrate binding. Related references: PMID:33571455, PMID:30237167
- ABHD12 also hydrolyzes oxidized phosphatidylserine, a proapoptotic "eat me" signal produced during severe inflammatory stress, linking it to apoptotic cell clearance. Related references: PMID:30643283
- Subcellularly, ABHD12 is localized at the endoplasmic reticulum membrane, where it exerts its lipase functions. It is a transmembrane glycoprotein, and alternative splicing generates multiple isoforms.
- Mutations in ABHD12 are associated with PHARC syndrome (polyneuropathy, hearing loss, ataxia, retinitis pigmentosa, and cataract), underscoring its importance in neurological and sensory systems. The protein is linked to diseases such as deafness, neuropathy, and retinitis pigmentosa.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant fusion protein spanning amino acids 110–250 of human ABHD12 (NP_056415.1). This central region is designed to elicit antibodies recognizing the native protein in various applications.
- For Western blot (WB), a band at approximately 45 kDa (calculated MW 45,097 Da) is expected. Validate with positive control lysates from human or rat tissues/cell lines known to express ABHD12, such as brain.
- For ELISA, consider coating with the recombinant immunogen or a related peptide, and optimize antibody dilution to achieve a linear detection range.
- Due to the antibody's cross-reactivity with human and rat, it can be used for both species; however, verify specificity in the biological context of interest.
